Protein Production Hands-On Workshop

This three-day course offers both foundational and practical training in recombinant protein production. Participants will gain hands-on experience across the entire workflow – from cell transformation, recombinant protein expression and purification to final sample concentration and purity validation.

Format

• Primarily hands-on tutorials, complemented by short lectures
• Participants will work in pairs to express and purify recombinant protein
• Lectures are open to external attendees upon prior consultation (due to seminar room capacity)
• Workshop capacity is limited; registration is required
• Tutorials and lectures will be led by technical scientists from the CMS Protein Production Core Facility:

Tuesday, November 3 – Protein Expression in E. coli (9:00 – 17:00)
09:00 – 09:20: Introduction (lecture)
10:00 – 10:30: Preparation for protein expression in 1 L E. coli culture (practical)
10:30 – 11:00: Cloning using restriction enzymes and RF cloning (lecture)
11:00 – 11:30: Overview of prokaryotic and eukaryotic expression systems we use (lecture)
12:00 – 14:00: OD measurements and induction of protein expression (practical)
14:00 – 16:00: E. coli transformation and group discussion
16:00 – 17:00: Chromatography techniques (lecture)

Wednesday, November 4 – Cell Harvesting & Initial Purification (9:00 – 16:00)
09:00 – 10:00: Cell harvesting (practical)
10:00 – 12:00: Sonication and centrifugation (practical)
13:00 – 16:00: Affinity chromatography, SDS-PAGE, dialysis (practical)
14:00 – 14:15: Media preparation (lecture)

Thursday, November 5 – Final Purification & Troubleshooting (9:00 – 15:00)
09:00 – 15:00: Protein concentration, Size Exclusion Chromatography, SDS-PAGE (practical)
